Big Red Isolator Screw on Hot Water Cylinder

I have a hot water cylinder upstairs which has two big red isolator screws, one at the top (goes directly into tank but also feeds off to a pump for shower) and one at the bottom (boiler return pipe?) on the pipework. Can anyone tell me what position these should be in, open or closed?
